چکیده:
زمینه و هدف: مواجهه با آرسنیک با احتمال آپوپتوز کبدی و نارساییهای متابولیک همراه است و احتمالا تمرین تناوبی شدید (HIIT) و کورکومین تاثیر مثبتی در این زمینه دارند. اما تاکنون تاثیر توام آنها بررسی نشده است. بنابراین هدف تحقیق بررسی تاثیر تمرین HIIT و کورکومین بر مقدار NRF2 و کاسپاز 3 کبد و همچنین ظرفیت تام ضداکسایشی (TAC)، گلوکز، تریگلیسرید (TG) و HDL خون در موشهای در معرض آرسنیک بود. روش تحقیق: در این تحقیق تجربی شش هفتهای، 48 موش نر به شش گروه شامل آرسنیک-HIIT (تمرین)، آرسنیک-کورکومین (کورکومین)، آرسنیک-کورکومین-تمرین (توام)، آرسنیک، کنترل اتانول و کنترل معمولی (کنترل) تقسیم شدند. آرسنیک روزانه با دوز mg/bw 5 و کورکومین روزانه mg/bw 15 از طریق گاواژ مصرف شدند و تمرینHIIT (پنج روز در هفته) در هر جلسه شامل 60 دقیقه فعالیت (تناوبهای چهار دقیقهای دویدن با شدت 90-85 درصد v VO2max، با دو دقیقه استراحت فعال) بود. دادهها با آزمون ANOVA با خطای کمتر از 05/0 تحلیل شدند. یافتهها: در گروه آرسنیک کاسپاز 3 کبد و همچنین گلوکز و TG خون بیشتر و TAC و HDL کمتر از گروه کنترل معمولی بود (001/0=P در همه موارد) که HDL، گلوکز و TAC در هر سه گروه تمرین (99/0=P، 56/0=P و 99/0=P)، کورکومین (99/0=P، 082/0=P و 66/0=P) و همچنین توام (99/0=P، 99/0=P و 060/0=P) و کاسپاز 3 کبد تنها در گروه توام (103=P)، تفاوت معنیداری با گروه کنترل معمولی نداشتند. NRF2 کبد فقط در گروههای کورکومین (001/0=P) و توام (001/0=P)، به بیش از گروه کنترل رسید. نتیجهگیری: آرسنیک کبد را در معرض استرس اکسایشی و بروز آپوپتوز قرار داده و موشها را با افزایش قند و چربی خون مواجه میکند. با اینکه تاثیر آرسنیک بر افزایش TG خون، توسط تمرین، کورکومین و یا اثر توام اصلاح نشد، ولی هر سه مداخله افزایش قند و کاهش HDL و TAC خون را اصلاح کردند. بااینحال، تنها اثر توام افزایش کاسپاز 3 کبدی ناشی از آرسنیک را اصلاح کرد و فقط کورکومین توانست مقدار NRF2 کبد را حتی به بیش از گروه کنترل برساند. ولی، به دلیل محدودیتهای تحقیق و کمبود شواهد، هنوز نیاز به تحقیقات بیشتر باقی است.
Aim & background: Arsenic exposure could likely lead to hepatic apoptosis and metabolic disturbances and high intensity interval training (HIIT) as well as curcumin supplementation seems to have beneficial effects, however, the concomitant effect of them has not still been evaluated. Therefore, the aim of this study was to investigate the effects of HIIT and curcumin supplementation on hepatic NRF2 and caspase 3 and blood total antioxidant capacity (TAC), glucose, triglyceride (TG) and HDL in male rats exposed to arsenic.Methods: In a six-week experimental study, 48 rats were randomized into six groups of arsenic-HIIT(HIIT), arsenic-curcumin(Curcumin), arsenic-HIIT+curcumin(Concomitant), Arsenic, Ethanol Control and normal control(Control). Arsenic 5 mg/bw.day and curcumin 15 mg/bw.day, were consumed orally during the entire study period. HIIT were conducted for six weeks (five d/w, 60 min/session, consisted of running bouts (four min) at 85-90% of v Vo2max with two min active rest intervals). The data were analyzed using one-way ANOVA with Results: Hepatic caspase 3 as well as circulatory glucose and TG were higher, while blood TAC and HDL level were lower in arsenic group compared to normal control (p=0.001 under any circumstances). Moreover, non-significant differences were observed in blood HDL, glucose and TAC levels, respectively in three groups of HIIT (P=0.99, P=0.56, P=0.99), Curcumin (P=0.99, P=0.082, P=0.66) and Concomitant (P=0.99, P=0.99, P=0.060) compared to Control group. Additionally, the liver caspase 3 level had non-significant difference with Control group just in concomitant group (P=0.010), while hepatic NRF2 were elevated to levels even higher than Control group in curcumin consuming (Curcumin and Concomitant) groups (p=0.001 under both circumstances).Conclusion: Arsenic renders the liver to oxidative stress and apoptosis and encounters the rats with both hyperglycemia and dyslipidemia. While the up-regulated circulatory TG Induced by arsenic could not restore in three intervention groups (HIIT, Curcumin and Concomitant), the elevated blood glucose and also the lowered HDL and TAC levels were efficiently restored. However, a higher hepatic caspase 3 was corrected just in Concomitant group with NRF2 elevation to levels even higher than Control group observed only by curcumin supplementation. However, more investigations still remain to be done because of the study limitations and lack of similar evidence in this area.
